Will most mods have to be redone after 2.1 or is 2.1 just a content patch?
That seems to be the most I can find out about 2.1 is new or updated content.

No, 2.1 does not have significant lua changes, mods will appear 'out of date' but generally still function.

From what I've seen, minor tweeking of many mods may be needed, but this isn't a 2.0.1 patch where all but the most basic of mod needed more work than just updating the API number.

Not to say that most mods won't get updated, 2.1 is bringing some interesting potential to some mods.

Like everyone else has said: No. There aren't significant changes. There are some minor things that may break some addons, but these things should be a very quick and easy fox for those broken addons.

QFT. There is going to be some great additions, but nothing breaking that I really foresee causing an uproar. Macros are getting some great additions as well. Heck, some of Blizzard's improvement to their own UI is going to negate the need for some addons entirely. New features such as loot window on cursor (YES!). And auto dismount when you click on abilities that you need to be dismounted on.

Trinity is borked on the PTR. Spamsentry will no longer work in it's current form.

These are the only two I have read about.
